The short version of Enkephalinase fits in a sentence. The long version — which is the one that helps — is below.
This page was last updated on 2025-10-31 and is reviewed periodically as new material appears.
Proposed mechanisms centre on modulation of the GABAergic system, with reports of altered expression of genes related to GABA-A receptor subunits and changed monoamine turnover. Some studies describe inhibition of enkephalinase, the enzyme that degrades endogenous enkephalins, which may prolong opioid peptide signalling. Effects on brain-derived neurotrophic factor and on cytokine expression have also been reported. These findings come largely from animal models and small human studies, and the precise primary target remains unresolved.
Published clinical evidence is limited. Most controlled trials were conducted in Russia, enrolled modest numbers of participants, and appeared in Russian-language journals, which restricts independent verification. Reported outcomes include lower anxiety scores, improved attention and memory measures, and changes in fatigue ratings. Reviews written in English note methodological limitations such as small samples and inconsistent endpoints. Whether the compound produces clinically meaningful benefit relative to established anxiolytics is therefore an open question rather than an established finding.
Lyophilized selank is normally supplied as a dry powder and is considered stable for extended periods when kept cold and dry. Moisture uptake is the main practical threat, because absorbed water promotes both hydrolysis and aggregation in the solid state. Vials are usually warmed to room temperature before opening so that condensation does not form on the powder. Supplier documentation commonly specifies -20 °C for routine storage, with -80 °C used for material intended to be archived for years.
Once dissolved, the peptide is markedly less stable than the dry powder. Aqueous solutions are subject to backbone hydrolysis and to microbial growth when they are handled without sterile technique. Buffered solutions near neutral pH are common for short-term laboratory work, while acidic conditions are sometimes used to improve solubility. Analytical laboratories generally prepare working solutions fresh rather than storing them, and a residual water film left in a reopened vial can seed degradation even when the container appears dry.

Purity assessment relies mainly on reverse-phase high-performance liquid chromatography with ultraviolet detection. Because the peptide lacks a strong chromophore, detection often uses backbone absorbance near 214 nm. Identity is confirmed by mass spectrometry, typically electrospray ionization or matrix-assisted laser desorption, comparing the measured mass against the expected value. Amino acid analysis can verify composition after acid hydrolysis. Diastereomer content and residual counterions are reported less often, although both can influence biological assays.
Lyophilized material is generally stable for extended periods when kept dry at or below minus twenty degrees Celsius. Working solutions are less stable, and common practice is to aliquot and freeze them so that repeated freeze-thaw cycles are avoided. Aqueous solutions are sensitive to pH extremes and to microbial growth, so short-term storage at refrigerator temperature is typical. Oxidation and hydrolysis are the principal degradation routes. Reconstitution with sterile water or a mild buffer is standard, and solutions should be protected from light.
Regulatory treatment varies by jurisdiction. In Russia the compound is a registered prescription product, while in the European Union and the United States it is generally handled as a research chemical without a marketing authorization. Suppliers therefore operate outside pharmaceutical oversight, and buyers rely on supplier documentation for purity and identity claims. Chain of custody and third-party testing are the main verification tools. Analysts note that the absence of a pharmacopoeial monograph for research-grade material limits standardization across vendors.
Animal studies have examined behaviour in tests of anxiety, memory retention and stress response, and several report changes in neurotrophic or neurotransmitter-related markers. The human evidence base is much smaller, consisting mainly of short trials conducted in Russia with limited reporting in English-language journals. Sample sizes are modest and outcome measures vary between studies, so the findings are best described as preliminary. Independent replication under modern trial standards has not been widely reported.
Outside its country of origin the compound is generally handled as a research chemical rather than an approved medicine. No regulatory approval from the United States Food and Drug Administration or the European Medicines Agency has been granted for human use. Identity and purity are normally checked by reverse-phase high-performance liquid chromatography, with mass spectrometry used to confirm the molecular mass. Lyophilised material is stored cold and desiccated, and repeated freeze-thaw cycles are avoided.
Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography, with mass spectrometry used to confirm molecular mass and sequence information. Amino acid analysis and peptide mapping may supplement these methods. Certified reference standards are scarce, and many commercial lots are sold as research chemicals without a pharmacopoeial monograph. Regulatory treatment differs by country: Selank is a registered prescription medicine in Russia, while in the European Union and the United States it is not an approved drug and may fall under research-chemical or unapproved-product frameworks.
Selank is a hydrophilic peptide and dissolves readily in water and in aqueous buffers. The lyophilised powder is typically a white to off-white solid. Because short peptides are prone to hydrolysis and oxidation, handling benefits from limiting exposure to heat, moisture and strong light. Working solutions are commonly prepared in sterile water or saline, and repeated freeze-thaw cycles are avoided to reduce aggregation and loss of activity. These practices reflect general laboratory convention rather than published stability specifications.
Dry powder is normally held at -20 degrees Celsius or lower, in a sealed container with desiccant and protection from light. Reconstituted solutions are usually kept at 2 to 8 degrees Celsius for short periods and frozen for longer ones. Proline residues at several positions are generally associated with some resistance to peptidase attack, but chemical stability still declines at neutral to alkaline pH and at elevated temperature. Exact shelf-life figures are product-specific and are not standardised across suppliers.
=== Buffer agents === By buffering any potential pH change, buffer agents can provide chemical stability and ensure the physical compatibility of the topical cream formulation. They ensure that the formulation can deliver the correct amount of drug to the therapeutic application site, is free from microbial contamination, and physically unchanged since the manufacturing day. Nonetheless, buffer agents need to be carefully added to avoid undesirable effects on physical stability. For example, buffer agents may influence the rheological behavior.
In the 1950s, Alfred Nier developed the technique of ID-TIMS, which later become the first tool used in monazite geochronology. Since this method involves the chemical separation of monazite (isotope dilution), it is regarded as a conventional analysis technique. Generally, it takes several hours for a U-Pb measurement. The precision of date is nearly 0.1%, provided that the ages are concordant (i.e. not dates reflecting mixing of zonations). It is regarded as the most precise method in monazite geochronology. Monazite mineral grains are carefully hand-picked for dating. They are spiked with a tracer solution and dissolved in HF or HCl. Using ion exchange chemistry, U, Th and Pb are separated from other elements. The purposes of the separation are (1) potential isobaric interference should be removed before analysis because of the high-sensitivity and low-mass resolution nature of TIMS; (2) ionization of the elements of interest maybe impeded by other elements, which results in reduced signal size and precision. The separated U, Th and Pb samples are put carefully onto a metal filament, which is usually made from Re. The elements are heated and ionize to their respective ions, which are accelerated under a strong magnetic field and are measured by a detector. The tracer solution is a solution with a known amount of U and Pb tracer isotopes. Due to elemental fractionation, both elements cannot be measured simultaneously by TIMS. The tracer solution is therefore used to measure ratios of sample isotope to tracer isotopes.

=== Wireless systems === Dry bells may have a through water communication system fitted as a backup. This is intended to provide communications in the event that the cable is damaged, or even if the bell is completely severed from the umbilical and deployment cables.
Pulmonary hypertension has been gaining recognition as a complication of chronic hereditary and acquired hemolysis. Free hemoglobin released during hemolysis inactivates the vasodilator nitric oxide (NO). Hemolysis also releases arginase that depletes L-arginine, the substrate needed for NO synthesis. This reduces NO-dependent vasodilation and induces platelet activation, thrombin generation, procoagulant factors and tissue factor activation, contributing to the formation of thrombosis. This can lead to esophageal spasm and dysphagia, abdominal pain, erectile dysfunction, systemic hypertension, decreased organ perfusion, promotion of inflammation and coagulation, and thrombosis. Chronic hemolysis may also lead to endothelial dysfunction, heightened endothelin-1-mediated responses and vasculopathy. The release of heme leads to the production of bilirubin and depletion of plasma proteins, such as albumin, haptoglobin, and hemopexin, which may lead to jaundice. It may also lead to increased levels of the heme breakdown product stercobilin in the stool. Splenectomy of those with hemolytic disorders appears to increase risk of developing pulmonary thrombosis. Complications may also arise from the increased workload for the kidney as it secretes erythropoietin to stimulate the bone marrow to produce more reticulocytes (red blood cell precursors) to compensate for the loss of red blood cells due to hemolysis.
=== Other Valve games === Valve developed several games using the GoldSrc engine, many of which were based on original user-made modifications. Valve's Team Fortress Classic, released in 1999, was developed primarily by two of the developers of the Quake mod Team Fortress. Counter-Strike and Day of Defeat were also originally Half-Life modifications that Valve purchased the rights to and re-released as standalone titles. Counter-Strike evolved into its own series with the debut of the Japanese arcade game Counter-Strike Neo in 2003 and Valve's own follow-up in 2004, Counter-Strike: Condition Zero, both of which run on the GoldSrc engine. Although Valve's further installments in the series starting with Counter-Strike: Source use the newer Source engine instead, Counter-Strike Online and Counter-Strike Nexon, two spinoff titles released by Nexon in 2008 and 2014 respectively, use GoldSrc as their basis.

== Biological activity == Prodigiosin received renewed attention for its wide range of biological activities, including activities as antimalarial, antifungal, immunosuppressant, and antibiotic agents. It is perhaps best known for its capacity to trigger apoptosis of malignant cancer cells. The exact mechanism of this inhibition is highly complex and not entirely elucidated, but could involve multiple processes, including phosphatase inhibition, copper mediated cleavage of double stranded DNA, or disrupting the pH gradient through transmembrane transport of H+ and Cl- ions. As a result, prodigiosin is a highly promising drug lead, and is currently in preclinical phase study for pancreatic cancer treatment. Prodigiosin has recently been found to have excellent activity against stationary phase Borrelia burgdorferi, the causative agent of Lyme disease. Prodigiosin has been also reported to exhibit antimicrobial activity against Gram-positive bacteria including methicillin resistant Staphylococcus aureus (MRSA), Staphylococcus aureus, Enterococcus faecalis, as well as Gram-negative Escherichia coli, Vibrio vulnificus, Salmonella enterica. Although the exact mechanism of action is not completely understood, recent molecular docking studies show that prodigiosin may interact with fatty acid biosynthetic enzyme in bacteria. Prodigiosin is also found to act as algaecide against harmful algal bloom.
== Peptide nanodisc == In peptide nanodiscs, the lipid bilayer is screened by amphipathic peptides instead of two MSPs. Peptide nanodiscs are structurally similar to MSP nanodiscs and the peptides also align in a double belt. They can stabilise membrane proteins, but have higher polydispersity and are structurally less stable than MSP nanodiscs. Recent studies, however, showed that dimerization and polymerization of the peptides make them more stable.
Mono-N-protected amino acid (MPAA) is a bifunctional ligand that plays a key role in C–H functionalizations by accelerating the reaction rate and imparting specified chirality into the product. Amino acids are ideal building blocks for chiral ligand synthesis due to the cost, accessibility, large variety, solubility, and inherent chirality. Naturally occurring amino acids are transformed into chiral MPAA ligands that, upon coordination to metal complexes, allow reactions to occur that are otherwise energetically unfavorable. Great strides in the development of MPAA ligands over the past two decades have led to the integral role that enantioselective catalysis now plays in complex organic synthesis.
== Mechanism of action == Larazotide is an inhibitor of paracellular permeability. In celiac disease, one pathway that allows fragments of gliadin protein to get past the intestinal epithelium and subsequently trigger an immune response begins with binding of indigestible gliadin fragments to the chemokine CXC motif receptor 3 (CXCR3) on the luminal side of the intestinal epithelium (see this page). This leads to the induction of myeloid differentiation factor 88 (MYD88) and the release of zonulin into the lumen. Zonulin then binds to epidermal growth factor receptor (EGFR) and protease-activated receptor 2 (PAR2) in the intestinal epithelium. This complex then initiates a signalling pathway that eventually results in tight junction disassembly and increased intestinal permeability. Larazotide acetate intervenes in the middle of this pathway by blocking zonulin receptors, thereby preventing tight junction disassembly and associated increase in intestinal permeability.

It is a seven-amino-acid peptide, Thr-Lys-Pro-Arg-Pro-Gly-Pro, produced by chemical synthesis rather than extracted from biological tissue. Its design is based on tuftsin, a natural immunomodulatory tetrapeptide. The C-terminal Pro-Gly-Pro segment is a common stabilising motif in short regulatory peptides.
The core four residues correspond to tuftsin, which occurs naturally as part of immunoglobulin G. The full seven-residue sequence, however, is not a known endogenous peptide. It is a laboratory-designed analogue intended to combine tuftsin-like activity with greater resistance to breakdown.
No single receptor has been confirmed as the primary target. Reports describe involvement of the GABAergic system, interference with enkephalin degradation, and shifts in neurotrophic factor expression. Because these observations come from different models and assays, they have not yet been integrated into one accepted mechanism.
Sealed, desiccated storage at -20 °C or colder is the standard recommendation for research-grade material. Vials should reach room temperature before they are opened, which limits condensation. Repeated temperature cycling is discouraged.
