This is a working overview of RP-HPLC, written for readers who want more than a one-paragraph summary but less than a textbook.

Peptide stability depends strongly on temperature, moisture, and pH. Lyophilized Selank is generally most stable when stored cold and dry, with freezer temperatures commonly used for long-term storage. In solution, the compound is susceptible to hydrolysis and to microbial growth if it is not handled aseptically. The C-terminal proline-rich extension appears to slow enzymatic cleavage relative to tuftsin, though quantitative degradation rates vary with the matrix and the conditions tested. Published stability data specific to Selank remain sparse.
Quality assessment of Selank samples typically combines purity determination with identity confirmation and counter-ion analysis. Purity is usually reported as a percentage by chromatographic area, with values above 95 percent often quoted for research-grade material. Water content and residual solvents are checked in lyophilized batches because they affect both stability and accurate mass determination. A reported purity figure does not by itself establish that a sample is the intended sequence, so orthogonal methods are needed to rule out sequence isomers or truncation products.
Handling follows standard practice for research peptides. Material is weighed in a low-humidity environment because the powder absorbs atmospheric moisture. Purity is reported as the percentage area of the main peak in a chromatogram, with specifications commonly set at 95 percent or higher; values below that threshold indicate the presence of truncated or modified species. Residual trifluoroacetate from purification is often present and may affect mass balance. Certificates of analysis should state the analytical method, the column and gradient used, and the lot-specific retention time so that results can be compared across suppliers.
Identity and purity of selank are established with reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The peptide elutes from C18 columns with acetonitrile gradients in water containing trifluoroacetic acid or formic acid, and detection is usually performed by ultraviolet absorbance near 214 nm. Electrospray ionization in positive mode gives a doubly protonated ion near m/z 377, consistent with a mass of about 752 Da. Amino acid analysis or tandem mass spectrometry of fragment ions confirms the sequence. Because the molecule has no aromatic residues, it lacks a usable 280 nm chromophore, so low-wavelength detection or mass spectrometry is required.
Peptide bonds in selank are susceptible to hydrolysis under strongly acidic or basic conditions, and the terminal proline residues are vulnerable to exopeptidase activity in biological samples. Lyophilized powder stored dry at -20 °C typically remains stable for extended periods, whereas aqueous solutions degrade faster and may lose measurable purity within days to weeks depending on pH, temperature, and microbial load. Repeated freeze-thaw cycles promote aggregation and adsorption to container surfaces. For analytical work, solutions are usually prepared fresh, kept cold, and used within a single working day.

Published clinical evidence is limited. Most controlled trials were conducted in Russia, enrolled modest numbers of participants, and appeared in Russian-language journals, which restricts independent verification. Reported outcomes include lower anxiety scores, improved attention and memory measures, and changes in fatigue ratings. Reviews written in English note methodological limitations such as small samples and inconsistent endpoints. Whether the compound produces clinically meaningful benefit relative to established anxiolytics is therefore an open question rather than an established finding.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a structural analogue of tuftsin, a naturally occurring tetrapeptide fragment of the immunoglobulin heavy chain. The added Pro-Gly-Pro tail was intended to slow enzymatic degradation and extend biological activity. In Russia it is registered as an anxiolytic nasal preparation, while regulators elsewhere have not approved it for clinical use.
Naming conventions place Selank in the same research family as Semax, another Russian-developed peptide investigated for cognitive effects. The two compounds share a lineage but differ in sequence and in the biological systems proposed as their targets. Semax descends from ACTH fragments, whereas Selank descends from tuftsin. Publications sometimes identify Selank by its full peptide sequence or by laboratory codes rather than one uniform trade name. Because replication outside Russia is limited, reports on its properties are best read alongside the study design and the purity of the material tested.
Selank is a synthetic heptapeptide developed in Russia during the 1990s. Researchers at the Institute of Molecular Genetics of the Russian Academy of Sciences designed it as a stabilized analog of tuftsin, a naturally occurring immunomodulatory tetrapeptide. The compound has been studied primarily for its reported anxiolytic and nootropic effects. It remains largely unknown in Western pharmacology and is not approved as a medicine by major regulators such as the FDA or the EMA.
The primary structure of Selank is Thr-Lys-Pro-Arg-Pro-Gly-Pro, corresponding to the molecular formula C33H57N11O9 and a monoisotopic mass of roughly 751.9 daltons. The N-terminal threonine and the arginine residue in the fourth position are shared with tuftsin, which carries the sequence Thr-Lys-Pro-Arg. The three additional residues at the C-terminus, Pro-Gly-Pro, extend the chain and are associated with greater resistance to enzymatic degradation. This extension also separates Selank from the shorter parent peptide.
Lee & Co as concrete contractor using a Lee's Mixer, an early example of concrete mixers used during construction on the Admiralty Pier. Further construction from 1897 onwards established the Eastern Arm of the current harbour, the Southern Breakwater and a further extension of the Admiralty Pier. During the First World War, a former cargo ship, the Spanish Prince, was deliberately sunk by the Admiralty close to the entrance to Dover Harbour to prevent German U-Boats from firing torpedoes into the harbour. During World War II, the remains of another ship, the War Sepoy, were sunk alongside the Spanish Prince.

Pilots with gas utilities and biogas plants are underway with companies like Modern Hydrogen. Volume production is also being evaluated in the BASF "methane pyrolysis at scale" pilot plant, the chemical engineering team at University of California - Santa Barbara and in such research laboratories as Karlsruhe Liquid-metal Laboratory (KALLA). Power for process heat consumed is only one-seventh of the power consumed in the water electrolysis method for producing hydrogen. The Australian company Hazer Group was founded in 2010 to commercialise technology originally developed at the University of Western Australia. The company was listed on the ASX in December 2015. It is completing a commercial demonstration project to produce renewable hydrogen and graphite from wastewater and iron ore as a process catalyst use technology created by the University of Western Australia (UWA). The Commercial Demonstration Plant project is an Australian first, and expected to produce around 100 tonnes of fuel-grade hydrogen and 380 tonnes of graphite each year starting in 2023. It was scheduled to commence in 2022. "10 December 2021: Hazer Group (ASX: HZR) regret to advise that there has been a delay to the completion of the fabrication of the reactor for the Hazer Commercial Demonstration Project (CDP).

=== Colloquialisms === A large central adiposity deposit has been assigned many common use names, including "spare tire", "love handle", "paunch", "boiler", and "potbelly". Several colloquial terms used to refer to central obesity, and to people who have it, refer to beer drinking. However, there is little scientific evidence that beer drinkers are more prone to central obesity, despite its being known colloquially as "beer belly", "beer gut", or "beer pot". One of the few studies conducted on the subject did not find that beer drinkers are more prone to central obesity than nondrinkers or drinkers of wine or spirits. Chronic alcoholism can lead to cirrhosis, symptoms of which include gynecomastia (enlarged breasts) and ascites (abdominal fluid). These symptoms can suggest the appearance of central obesity.
==== Hydrodynamic countercurrent chromatography (CCC) ==== The operating principle of CCC instrument requires a column consisting of an open tube coiled around a bobbin. The bobbin is rotated in a double-axis gyratory motion (a cardioid), which causes a variable gravity (G) field to act on the column during each rotation. This motion causes the column to see one partitioning step per revolution and components of the sample separate in the column due to their partitioning coefficient between the two immiscible liquid phases used. There are many types of CCC available today. These include HSCCC (High Speed CCC) and HPCCC (High Performance CCC). HPCCC is the latest and best-performing version of the instrumentation available currently.
Nanoparticles made from natural polymers that are biodegradable have the abilities to target specific organs and tissues in the body, to carry DNA for gene therapy, and to deliver larger molecules such as proteins, peptides, and even genes. To manufacture these polymeric nanoparticles, the drug molecules are first dissolved and then encapsulated or attached to a polymer nanoparticle matrix. Three different structures can then be obtained from this process; nanoparticles, nanocapsules (in which the drug is encapsulated and surrounded by the polymer matrix), and nanospheres (in which the drug is dispersed throughout the polymeric matrix in a spherical form). One of the most important traits for nanoparticle delivery systems is that they must be biodegradable on the scale of a few days. A few common polymer materials used for drug delivery studies are polybutyl cyanoacrylate (PBCA), poly(isohexyl cyanoacrylate) (PIHCA), polylactic acid (PLA), or polylactide-co-glycolide (PLGA). Human serum albumin (HSA) and chitosan are also materials of interest. PBCA undergoes degradation through enzymatic cleavage of its ester bond on the alkyl side chain to produce water-soluble byproducts. PBCA also proves to be the fastest biodegradable material, with studies showing 80% reduction after 24 hours post intravenous therapy injection. PIHCA, however, was recently found to display an even lower degradation rate, which in turn further decreases toxicity.
==== Shotgun sequencing ==== High throughput shotgun sequencing using tools such as Solexa or Illumina yields approximately 5 million sequence tags per sample of maternal serum. Aneuploid pregnancies such as trisomy were identified when testing at the fourteenth week of gestation. Fetal whole of genome mapping by parental haplotype analysis was completed using sequencing of cffDNA from maternal serum. Pregnant females were studied using a 2-plex massively parallel maternal plasma DNA sequencing and trisomy was diagnosed with z-score greater than 3. The sequencing gave sensitivity of 100 percent, specificity of 97.9 percent, a positive predictive value of 96.6 percent and a negative predictive value of 100 percent.
=== Pharmacodynamics === AM-1235 is a cannabinoid receptor agonist with Ki of 1.5 nM at CB1 compared to 20.4 nM at CB2. While the 6-nitro substitution on the indole ring reduces affinity for both CB1 and CB2 relative to the unsubstituted parent compound AM-2201, CB2 affinity is reduced much more, resulting in a CB1 selectivity of around 13 times. This is in contrast to other related compounds such as AM-1221 where a 6-nitro substitution instead confers significant selectivity for CB2.

Gill recommended that people should "eat here at least once to find out what is really going on in your mouth". Also in 2005, the German critic Wolfram Siebeck visited the restaurant and complained of the delays in service and of several of the dishes. He described the mustard ice cream in a red cabbage gazpacho soup as a "fart of nothingness". The chef Nico Ladenis said, "Someone who makes egg and bacon ice cream is hailed a genius. If you vomit and make ice cream out of it, are you a star?" Tony Naylor of The Guardian enjoyed his trip in 2008. He defended the price, likening the occasional purchase of an exceptional meal to spending on entertainment or hobbies. In 2012, Fodor's described the Fat Duck as "extraordinary" and "one of the best restaurants in the country", and Frommer's gave it three stars, grading it "exceptional". In 2005, the Fat Duck was ranked first on the list of the World's 50 Best Restaurants. After spending 11 years on the list, it dropped to 73rd. It has been ranked second-best on numerous occasions, first behind the French Laundry and then behind El Bulli. In 2012, it was ranked 13th. In 2009, the Fat Duck was the only restaurant to be given a top score of ten out of ten in the Good Food Guide. The editor of the guide, Elizabeth Carter, wrote: "It's extremely rare that a restaurant cooks perfectly on a consistent basis, but we've had so many superlative reports that we're delighted to recognise the Fat Duck as the best restaurant in Britain." It retained that score through to the 2013 edition.
=== Temperature Tuesday === In 2010, Runza introduced their Temperature Tuesdays promotion. In January and February, the coldest temperature at a Runza at 6AM would be the price of an original Runza sandwich with the purchase of medium french fries and a drink (ex. if the coldest temperature at a Runza was 11° at Ogallala, an original Runza would be 11¢ with purchase of a medium fry and drink). If the temperature was below zero, a Runza would be free with the purchase of a medium fry and drink. The promotion has since been shortened to only take place in January.

== Career == Harrington was a programmer at the video game developer Dynamix and a designer on the Windows NT operating system at Microsoft. In 1996, he founded Valve with Gabe Newell, another former Microsoft employee. Harrington sold his Microsoft shares to fund his half of the startup. His wife at the time, Monica Harrington, was Valve's marketing strategist in its early years. Harrington worked as a programmer on Valve's first game, Half-Life (1998), and funded its development with Newell. It was a critical and commercial success. Harrington said: "At Microsoft you always wonder, 'Is it me being successful or is it Microsoft?' But with Half-Life I knew Gabe and I had built that product and company from scratch." On January 15, 2000, Harrington sold his stake in Valve to Newell and left to spend time with his wife. According to Newell, Harrington did not want to risk another project after the success of Half-Life. In 2006, Harrington co-founded the photo editing service Picnik with his friend and former colleague Darrin Massena. Picnik was acquired by Google in March 2010. Harrington left Google in March 2011. In January 2012, he co-founded another company with Massena, Catnip Labs. Harrington was CTO at the Committee for Children from 2016 to 2018, and CTO of Amplion from November 2018 until March 2020.
A carcinoid (also carcinoid tumor) is a slow-growing type of neuroendocrine tumor originating in the cells of the neuroendocrine system. In some cases, metastasis may occur. Carcinoid tumors of the midgut (jejunum, ileum, appendix, and cecum) are associated with carcinoid syndrome. Sometimes, carcinoids cause paraneoplastic syndromes, which involve discharge of serotonin and other vasoactive substances from well-differentiated carcinoids. A neuroendocrine paraneoplastic syndrome involves neoplastic secretion of functional peptides, hormones, cytokines, growth factors, and/or immune cross-reactivity between tumor tissues and normal host tissues, resulting in a syndrome of clinical signs and symptoms. Carcinoid tumors are the most common malignant tumor of the appendix, but they are most commonly associated with the small intestine, and they can also be found in the rectum and stomach. They are known to grow in the liver, but this finding is usually a manifestation of metastatic disease from a primary carcinoid occurring elsewhere in the body. They have a very slow growth rate compared to most malignant tumors. The median age at diagnosis for all patients with neuroendocrine tumors is 63 years.

The most common approach combines reverse-phase liquid chromatography with mass spectrometry. Chromatography separates the components while mass spectrometry confirms the molecular mass. Peptide sequencing or tandem mass analysis can further verify the amino acid order.
Lyophilized powder is usually kept frozen and protected from moisture. Solutions are less stable and are often used promptly or divided into aliquots to avoid repeated freeze-thaw cycles. Exact shelf-life values depend on purity and handling.
No. A purity figure derived from chromatographic area does not prove the amino acid sequence. Identity requires an orthogonal method such as mass spectrometry or sequencing. Truncated or isomerized peptides can co-elute with the target compound.
Identity is confirmed by matching the retention time in reversed-phase chromatography against a reference standard and by measuring the molecular mass with mass spectrometry. Tandem mass spectrometry or amino acid analysis can verify the sequence of the seven residues. Because the peptide contains no aromatic amino acids, detection at 280 nm is not useful.
