== Mechanism of action == Colistin is a polycationic peptide and has both hydrophilic and lipophilic moieties. These cationic regions interact with the bacterial outer membrane by displacing magnesium and calcium bacterial counter ions in the lipopolysaccharide. The hydrophobic and hydrophilic regions interact with the cytoplasmic membrane just like a detergent, solubilizing the membrane in an aqueous environment. This effect is bactericidal even in an isosmolar environment. Colistin binds to lipopolysaccharides and phospholipids in the outer cell membrane of Gram-negative bacteria. It competitively displaces divalent cations (Ca2+ and Mg2+) from the phosphate groups of membrane lipids, which leads to disruption of the outer cell membrane, leakage of intracellular contents and bacterial death. Colistin has also been reported to target tubulin, favorizing its polymerization.
Melanotan-II may cause reversible darkening of moles and freckles. It is unclear if Melanotan II can increase (or reduce) the risk of developing melanoma. Reports of melanomas associated with the use of Melanotan II have coincided with heavy sunbathing and sun bed use. A 2013 scientific review found there was no conclusive evidence it causes melanoma, and a 2021 review concluded "the increased risk of melanoma in Melanotan users, who use it for tanning and exhibit sun-seeking behaviour, can probably be explained by more UV exposure". === 1970: In the Wake of Poseidon and Lizard === King Crimson spent 1970 in a state of flux with various line-up changes, thwarted tour plans, and difficulties in finding a satisfactory musical direction, while Fripp was learning and developing as a songwriter during the writing process of the next three albums. As well as guitar, Fripp took on keyboard duties, while Sinfield expanded his creative role to operating synthesizers. Following McDonald's and Giles's departure, Lake, unsure of the band's future without them, began discussions with Keith Emerson of the Nice about possibly forming a new band together. With Fripp and Sinfield planning the recording of the second King Crimson album, and Lake's position uncertain, the band's management booked Elton John to sing on the album, a decision cancelled by Fripp after he listened to John's then-only album, Empty Sky, and deemed his singing style to be not right for King Crimson. Lake agreed to stay with the band until Emerson had completed remaining commitments with the Nice, at which point he left to form Emerson, Lake & Palmer with Emerson and Carl Palmer of Atomic Rooster. On the resulting In the Wake of Poseidon album, Lake provided all the lead vocals except on "Cadence and Cascade", which featured Fripp's old school friend Gordon Haskell. The sessions also included Michael and Peter Giles on drums and bass respectively, saxophonist Mel Collins (formerly of the band Circus), and jazz pianist Keith Tippett. === Differential diagnosis === Due to the comparative rarity of NMS, it is often overlooked. Immediate treatment for the syndrome should not be delayed as it has a high mortality of between 10-20%. Differentiating NMS from other neurological disorders can be very difficult. The diagnosis is suggested on patients with a history of drug exposure to the most common inducing agents such as strong antidopaminergic medications. The differential diagnosis includes serotonin syndrome, encephalitis, toxic encephalopathy, status epilepticus, heat stroke, catatonia and malignant hyperthermia. Substance intoxication from stimulant drugs, such as cocaine, amphetamine, or methamphetamine may also produce similar symptoms. gene expression The set of processes by which the information encoded in a gene is used in the synthesis of a gene product, such as a protein or a non-coding RNA, or otherwise made available to influence one or more phenotypes; both the product and the gene encoding it are then said to be expressed. Canonically, the first step is transcription, which produces a messenger RNA molecule complementary to the DNA molecule in which the gene is encoded. For protein-coding genes, the second step is translation, in which the messenger RNA is read by a ribosome to produce a polypeptide and ultimately a protein. Sanger's reagent proved effective at labelling the N-terminal amino group at one end of the polypeptide chain. He then partially hydrolysed the insulin into short peptides, either with hydrochloric acid or using an enzyme such as trypsin. The mixture of peptides was fractionated in two dimensions on a sheet of filter paper, first by electrophoresis in one dimension and then, perpendicular to that, by chromatography in the other. The different peptide fragments of insulin, detected with ninhydrin, moved to different positions on the paper, creating a distinct pattern that Sanger called "fingerprints". The peptide from the N-terminus could be recognised by the yellow colour imparted by the FDNB label and the identity of the labelled amino acid at the end of the peptide determined by complete acid hydrolysis and discovering which dinitrophenyl-amino acid was there. By repeating this type of procedure Sanger was able to determine the sequences of the many peptides generated using different methods for the initial partial hydrolysis. These could then be assembled into the longer sequences to deduce the complete structure of insulin. Finally, because the A and B chains are physiologically inactive without the three linking disulfide bonds (two interchain, one intrachain on A), Sanger and coworkers determined their assignments in 1955. Sanger's principal conclusion was that the two polypeptide chains of the protein insulin had precise amino acid sequences and, by extension, that every protein had a unique sequence.
